Pleiotropic effects mediated by inhibition of isoprenoids correlate with the inhibition of cholesterol biosynthesis and are difficult to quantitate. Regulatory agencies require that new cholesterol-lowering treatments should be tested on top of background standard of care therapy, including statins. The most important positive effects of statins are anti-inflammatory, anti-proliferative, antioxidant, immunomodulatory, neuroprotective, anti-diabetes, and antithrombotic, improving endothelial dysfunction and attenuating vascular remodeling besides many others which are discussed under the scope of this review. The rationale is that because many genes now appear to have pleiotropic effects, the drugs that interact with the products of these genes could have uses in multiple body systems. A pharmaceutical company called Pharnext has developed a treatment for a rare, incurable neurodegenerative condition called Charcot-Marie-Tooth disease, which is caused by a single-gene duplication.
